M2139 binds to the triple helical J1 epitope on collagen II. Binding to collagen II epitopes was determined in bead-based flow immunoassay as median fluorescence intensity (MFI). Triple helical (Gly-Pro-Hyp)x11 is a synthetic control peptide with 11 repeats of the Gly-Pro-Hyp sequence and mimics the collagen II backbone structure.


Description

Affinity purified mouse monoclonal anti collagen II (COL2A1) antibody clone M2139 in PBS, sterile filtered (0.2 µm). The antibody recognizes the J1 epitope on triple helical collagen II1. The J1 epitope on mouse collagen consists of residues 551–564 with sequence MPGERRGAAGIAGPK1,4. The antibody M2139 has been used in applications including immunohistochemistry4, Western blot2, immuno­precipitation3, and in vivo4 and in vitro5 functional assays.

Q1: Why choose M2139 over polyclonal anti-collagen II antibodies?

  • M2139 is a monoclonal antibody that binds a single, well-defined epitope (J1: residues 551–564) on the triple-helical region of native collagen type II. This ensures minimal cross-reactivity with other collagen types (e.g., I, III, XI), unlike polyclonals which may bind multiple epitopes across different collagens. Monoclonal antibody M2139 works  consistently from batch-to-batch and is conformation sensitive: M2139 recognizes the native triple-helical form, making it ideal for detecting intact collagen II in tissue sections or functional assays.

Q2: Which secondary antibody should I use with M2139?

  • M2139 is a mouse IgG2b isotype and an anti-mouse IgG2b or anti-mouse IgG  work as a secondary antibody. Choose the conjugate for your application. For  example, for immunohistochemistry (IHC) an anti-mouse HRP- or AP conjugate is appropriate. For immunofluorescence (IF) and flow cytometry (FCM) an anti-mouse IgG2b fluorophore-conjugate chosen based on your setup is the choice. 

Q3: Can M2139 induce arthritis in experimental models?

  • Yes. M2139 has been used to induce collagen antibody-induced arthritis (CAIA) in mice when co-administered with lipopolysaccharide (LPS), making it valuable for autoimmune arthritis research. M2139 is a component of Cab3 and Cab4 cocktails of cartilage-binding antibodies that initiate joint inflammation in preclinical models  of arthritis (PMID: 32448385)

Q4: How does M2139 compare to other monoclonals like CIIC1 or 5B2?

  • M2139’s defined epitope, arthritis-inducing capability, and low cross-reactivity make it a gold standard for native collagen II detection and autoimmune arthritis models. M2139 binds to the J1 epitope regardless of citrullination , whereas binding of anti-collagen antibody CIIC1 may be abolished by double citrullination. M2139 binds to the well-defined J1 epitope on COL2A1, whereas the 5B2 binds the N-terminal telopeptide (PMID: 7687475).
