The mouse Anti-citrullinated COL2A1 Antibody clone ACC4 binds to the C1 epitope on alpha-chain of collagen II when the first arginine in the epitope (position 359–369; ARGLTGRPGDA) is citrullinated (Cit-Arg). The antibody does not bind to the C1 epitope when the second arginine is citrullinated (Arg-Cit), neither when both arginines are citrullinated (Cit-Cit). The antibody binds to the epitope in both triple helical and cyclic form. Binding to collagen II epitopes was determined in bead-based flow immunoassay as median fluorescence intensity (MFI).


Description

Affinity purified mouse monoclonal anti collagen II (COL2A1) antibody clone ACC4 in PBS, sterile filtered (0,2 µm). The ACC4 antibody is an anti-citrullinated protein antibody (ACPA) that on COL2A1 binds specifically to citrullinated C1 epitope on alpha-chain. This antibody has been used in applications including immunohistochemistry (IHC) and in vivo functional studies such as collagen antibody induced arthritis and evoked pain-like behavior study12.

Immunogen

The antibody was generated by immunization of DBA/1 or (B10.Q × DBA/1) F1 mice with Protein-arginine deiminase type-4 (PAD4) -treated triple helical CII peptides containing T and B cell epitopes2.

Epitope

The ACC4 antibody is specific to citrullinated C1 epitope (position 359–369; ARGLTGRPGDA) on type 2 collagen alpha chain.

Q1: What epitopes does ACC4 bind?

  • ACC4, as a murine anti-citrullinated protein antibody (ACPA), can interact with one specific citrullinated epitope on collagen type 2 (COL2). Due to its private-specific nature, ACC4 binds only to the denatured/cyclic exposed citrullinated C1 epitope on COL2. The binding is conformational, thus it does not interact with the triple helical structure of C1 epitope.

Q2: Can ACC4 induce experimental arthritis?

  • ACC4 has been characterized in animal models, one typical utilization is for collagen antibody induced arthritis (CAIA). However, ACC4 alone cannot induce CAIA, it is usually combined with other monoclonal antibodies (e.g. M2139) as a cocktail to enhance the arthritogenic potency.

Q3: Which assays can ACC4 be used for?

  • Taking the advantage of the well characterized epitope specificity, the antibody is highly flexible for adaptation in different purposes. The most common assays are ELISA and immunohistochemistry (IHC), where the antibody could be used for capturing denatured C1 epitope on COL2, or denatured citrullinated COL2 protein, which is usually present in arthritic cartilage. It does not bind to psoriatic skin or esophagus tissue, which can be optimized as a control for Vacara's ACC1 antibody.

Q4: Which secondary antibody should I choose for ACC4?

  • The isotype of ACC4 is mouse IgG2b or IgG1 (please contact us for other isotypes). We have tested the antibody in multiple assays and can confirm that conventional anti-mouse IgG products are ideal as a secondary antibody. Optionally, anti-mouse kappa secondary antibody also performs similarly well.
